POTENSI SARI BIJI KEDELAI ( Glycine max), RIMPANG JAHE (Zingiber officinale) DAN KOMBINASINYA TERHADAP KADAR MDA DAN SOD JANTUNG TIKUS MODEL DIABETES
Abstract
Introduction: Hyperglycemia in diabetes mellitus (DM) contributes to the complications of diabetic cardiomyopathy. Soybean seeds (Glycine max) and ginger rhizome (Zingiber officinale) are efficacious herbs as antidiabetic and their complications, but research on the combination of both is still rare. This study aims to prove the effect of soybean seeds, ginger rhizomes, and their combination on MDA and SOD cardiac levels in diabetic rat.
Methods: This study used Sprague dawley male rats aged 12 weeks, weight 180-200 grams divided into 2 control groups and 4 test groups (n = 5 animals). The experimental animals were induced with a high-fat-fructose (DTLF) and streptozotocin 25 mg / kgBB intraperitoneal (i.p) multiple dose diet. Subsequently given soybean seed extract 5 g / kg BB, ginger rhizome extract 500 mg / kg BB, combination of soybean-ginger (delja) 1 2500: 250 mg / kg BB and combination of delja 2 5000: 500 mg / kg BW given orally for 30 days. Cardiac malondialdehyde levels were measured using MDA rat kit and cardiac SOD levels were measured using SOD rat kit. Data analysis using One Way Anova followed by LSD or Tamhane test (p <0.05).
Results: The administration of soybean seed extract, ginger rhizome, combination of delja 1 and 2 reduced cardiac MDA levels in a row around 80%, 15%, 30% and 55% compared to the control treatment group (p <0.05). The cardiac SOD extracts of soybean extract, ginger rhizomes, and combination of delja 2 increased by approximately 30%, 55% and 2% respectively, while the combination of delja 1 actually reduced SOD by about 15% compared to the control treatment group (p <0.05). While the effect of giving DTLF and STZ in the treatment control group increased cardiac MDA levels by about 5-fold and reduced cardiac SOD levels by approximately 20% compared to the normal control group (p <0.05).
Conclusion: The administration of soybean seed extract, ginger rhizome and its combination reduced levels of cardiac MDA in DM rat. Meanwhile, the administration of soybean and ginger rhizome extract inhibited the decrease levels of cardiac SOD in DM rat.
