Efek Antibakteri Kombinasi Ekstrak Metanolik atau Dekokta Daun Sirsak (Annona muricata L) dengan Amoksisilin Pada Bakteri Staphylococcus aureus atau Escherichia coli secara In Vitro

Abstract


Introduction:Antibiotic resistance is now aglobal problem. Alternative strategy to prevent antibiotic resistance is using herbs as anantibiotic adjuvant.Annona muricata L leaf extracthas an antibacterial activity, so it ispotential to be antibiotic adjuvants. This research aim to know the interaction of combination betweenA. muricataL leaf extract and Amoxicillin on S. aureus and E. coli.

Method:Extraction method was used metanolic extraction and decoction.Zone of inhibition was evaluatedwithKirby Bauer (disc)test, and their interaction were calculated withAmeri-Ziaei Double Antibiotic Synergism Test (AZDAST). Data were concidered significant atp<0,05. The variables were divided into 5 groups: high and low dosage of amoxicillin (HDA and LDA), high dosage of leaf A.muricata L methanolic extract (HDHM),highdosagesleaf of A.muricataL decoction (HDHD), methanolicextract of A.muricata and amoxicillincombination (EMAA), and leafA.muricataLdecoction-amoxicillin (HDTD) compound in S. aureusand E. coli.

Results:Zone of inhibition on S. aureus from low dosage antibiotic (ADR) and high dosageA.muricataLmethanolic extract (HDTM, 14,0±2,1 mm) werehigher in comparisen with single ADR (8,0±0,0mm, p0,037). In other hand, high dosageA.muricata Ldecoction extract(HDTD, 21,0±0,6mm) were higher in comparison with single ADR (12,0±0,6mm, p0,045). There were no significant results on the combination of both decoction and methanolicextract ofA.muricata L with amoxicilin on E.coli

Conclusion: The combination high dosage of both decoction and methanolic extract of A.muricata L with low dosage amoxicillin showed synergistic effect onS. aureus.
