Efek Dekokta Daun Pulutan (Urena lobata) Terhadap Kadar Superoxide Dismutase (SOD) dan Malondialdehyde(MDA) Hepar Ikan Zebra (Danio rerio) Yang Dipapar Malathion Secara Kronik

Abstract


Background: Malathion is an organophosphate pesticide that is metabolized into malaoxon and free radicals. Malaoxon is acetylcholinesterase inhibitor and free radical cause condition of oxidative stress that affect levels of liver superoxide dismutase (SOD) and malondialdehydee (MDA) levels. Water extract of U. lobata leaf has antioxidant, anti-inflammatory and hepatoprotective potential. This study aims to prove the effect of U. lobata leaf water extract on SOD and MDA levels in zebra liver exposed to chronic malathion.

Methods: Experimental laboratory in vivo with a post test only control group design. Zebra fish were divided into 2 control groups and 3 groups giving water extract of U. lobata with dose 125 mg/l, 250 mg/l and 500 mg/l. SOD and MDA levels were measured using spectrophotometry with wavelengths 450 nm and 532 nm respectively.Data analysis using one way ANOVA followed by LSD with a significance level of p<0,05.

Result:Water extract ofU. lobata leaves at doses of 125 mg/l, 250 mg/l and 500 mg/linhibit decrease in liversuperoxide dismutase(SOD) levels by40%, 120% and80% (p<0,05) and inhibits the increasemalondialdehyde(MDA) levels of liver respectively40%, 59% and55%(p<0,05). The best dose of U. lobata therapy to inhibit decrease of SOD and inhibit increaseof MDA is 250 mg/l (p<0,05).

Conclusion: Water extract ofU. lobata leaves inhibit decreased insuperoxide dismutase (SOD) levels and inhibits the increase of Danio rerio’s liver malondialdehyde (MDA) levels which is exposed to chronically malathion 40 days.
