Pengaruh Berbagai Interval Pembatasan Waktu Makan pada Kadar Insulin Serum dan Jumlah Sel di Pulau Langerhans Pankreas Tikus Wistar Betina dengan Diet Atherogenik
Abstract
Introduction: Atherogenic diets can induced cell death in the islet of langerhans and caused insulin resistance leading to changes in insulin secretion. Time restricted feeding were known to regulate serum insulin levels and improve islet cells health. However, which type of time restricted feeding is best to regulate these effects is unknown.
Methods: Female Wistar rats (n=34) aged 5 weeks. Were divided into 5 groups treatment (n=6) KN,KP, KSH,KDH AND KSK. KN was given standard diet while KP was given atherogenic diet for 10 weeks. Group of restricted diet were daily fasted (KSH), one day interval (KDH) fasted, and Monday-Thursday (KSK) fasted with atherogenic diet for 4 weeks after 6 weeks of atherogenic diet without fasting. A pre-treatment group (KPP, n=4) was given atherogenic diet for 6 weeks to evaluated the state of pre-restricted treatment. At the end of treatment, serum insulin were measured using Rat Insulin ELISA Kit while the number of cell in the islet of Langerhans was manually count in histologic preparations. Result were analyzed and considered as statistically significant if p<0.05.
Results: Atherogenic diet tend to increase serum insulin levels and cell count. KSK restricted Feeding tend to have lower serum insulin levels compared to KDH or KSH. However this results were not statistically significant. Number of cell in the islet of langerhans in all treatment groups were 187.56±39.02, 208.03±32.57, dan 179.43±39.05 in KSK, KDH and KSH group respectively but data has no statistical significance. (p>0.05).
Conclusion: Daily, alternate and Monday-Thursday time restricted feeding have no stastical significance on the levels of serum insulin and cell count on the islet of Langerhans in female wistar Rats fed with atherogenic diets.
