UJI TOKSISITAS SUBKRONIK DEKOKTA DAUN NIMBA (Azadirachta indica A.Juss) PADA DOSIS MATC DAN LC50 TERHADAP JUMLAH HEPATOSIT NEKROSIS IKAN ZEBRA (Danio rerio) FASE JUVENIL
Abstract
Introduction : neem leaf has potential effect as an antidiabetic, antimalaria, and anticancer. However, some studies prove the negative effects of neem leaf, than still need to done a toxicity test for the LC50 value and the effect of neem leaf on the liver as a detoxification organ. Toxicity tests were performed on juvenile zebra fish. The purpose of this study was to know the LC50 dose, effect of the MATC and LC50 dose of Neem Leaf Decoctation (DDN) on the number of hepatocyte necrosis of juvenile zebra fish.
Methods: In vivo experimental laboratory research with control group post test only design to determine subchronic LC50 dose at 9 doses of DDN and calculation of hepatocyte necrosis in 4 groups (control, therapy, MATC, LC50) after being explained 15 days of juvenile zebrafish. LC50 data were analyzed using probit and number of hepatocyte necrosis were analyzed by Kruskal Wallis and Mann-Whitney, significant if p <0.05. Correlation test using spearman's rho.
Result: Subchronic LC50 value of DDN in juvenile zebrafish is 409,736mg / L. Therapy doses reduce hepatocyte necrosis (p> 0.05) 19.53% compared to control. MATC and LC50 doses increased hepatocyte necrosis (p <0.05) 82.5% and 84.4% compared to the control group and 85.9% and 87.4% compared to the therapy dose group.
Conclusion: Subcronic LC50 dose of the decocta of neem leaf is included in the category of almost toxic (409,736 mg/L). MATC and LC50 doses increases the number of hepatocyte necrosis of juvenile zebrafish
