Efek Ekstrak Infusa Daun Sirsak (Annona muricata Linn.) terhadap kadar TNF-α (tumor necrosis factor-alpha) Gaster Tikus Wistar Betina yang Diinduksi Indometasin
Abstract
Introduction : Non-steroidal Anti-Inflammatory Drugs (NSAID) are common anti-inflammatory drugs as anti-inflammatory, and as pain relief. NSAID have side effects increasing the risk of gastric ulcers due to COX-1 inhibition. Soursop leaves (Annona muricata Linn.) containing flavonoid, alkaloid, and tannin allegedly has an anti-inflammatory effect. This study aims to prove the effect of Infusion extract of soursop leaves on the levels of TNF-α rat gastric induced by indomethacin.
Method : Using the true experimental method with the control group design post test only in vivo, using 25 female wistar rats aged 8-10 weeks. Rats were divided into 5 groups consisting of Negative Control (KN), Positive Control (KP) Induced by Indomethacin and treatment groups (KP1, KP2, KP3) indomethacin induced and given Infusion extract of soursop leaves with a dose of 100 mg/KgBB , 200 mg/KgBB, and 400 mg/KgBB per sonde for 7 days. TNF-α levels of gastric tissue were measured by indirect ELISA method followed by reader ELISA with a wavelength of 450 nm. Data analysis using Kruskal wallis followed by Post Hoc Tamhane. The results are said to be significant if p <0.05.
Result : There were no significant differences in gastric TNF-α levels between KN and KP (p<0,05) and there were no significant differences in gastric TNF-α levels between KP and KP1, KP2, and KP3 (p>0.05). However, a tendency was found that increasing the dose of Infusion extract of soursop leaves would reduce TNF-α levels. The decrease in TNF-α levels in KP1 against KP was 14,83%, KP2 against KP was 15,94%, and KP3 against KP was 16,83%.
Conclusion : Infusion extract of soursop leaves at all doses did not reduce levels of TNF-α in rats.
