Potensi Anti Diabetes Fitosterol Biji Kedelai (Glycine max) [Studi in silico pada Glucose Transporter-4 (GLUT 4), Dipeptidyl Peptidase-4 (DPP-4), dan Alpha-glukosidase]
Abstract
Introduction: Soybean seeds (Glycine max) is one of the herbs that have the potential as an anti-diabetic, but the mechanism of action and phytosterols compounds that contribute not known with certainty. This study aims to prove the anti-diabetic potential of soybean seeds in inhibiting DPP-4 and α-glucosidase activity and increasing GLUT-4 activity through in silico studies.
Method: Soybean powder is extracted using the digestion method with ethanol as the solvent. Identification of active compounds in ethanol extract of Soybean Seeds using Liquid Chromatography- Tandem Mass Spectrometry (LC-MS/MS). The effect of active substances of Soybean Seeds on GLUT-4, DPP-4 and α-glucosidase were evaluated with in silico methode using web-based software (docking server) with Staurosporin, Vildagliptin, and Acarbose as the standard of comparison.
Result: The identified phytosterols of Soybean Seeds were β-Sitosterol, Campesterol, Stigmasterol, and Lanosterol. The in silico test by evaluating the value of free bonding energy (ΔG), inhibition constant (Ki) and surface interaction showed that Stigmasterol strongly inhibits DPP-4 and α-glucosidase activity and Lanosterol strongly bind to GLUT-4.
Conclusion: The In silico analysis of soybean seed active compounds that have potential as anti-diabetic with the mechanism of action inhibits DPP-4 and α-glucosidase activity and increases GLUT-4 activity is β-Sitosterol, Campesterol, Stigmasterol, and Lanosterol.
