EFEK KOMBINASI EKSTRAK INFUSA SAMBILOTO, SALAM, KAYU MANIS, DAN TEMULAWAK TERHADAP KADAR TNF-α SERUM TIKUS HIPERGLIKEMIA
Abstract
Introduction: The high incidence and complications due to hyperglycemia are associated with inflammation mediated by TNF-α. In pathologic condition, TNF-α worse the condition of insulin resistance and increases the risk of oxidative stres. Sambiloto, bay leaf, cinnamon, and ginger rhizome have antihyperglycemia, antioxidant and anti-inflammatory potency. This study aims to prove the effect of combined infusion sambiloto, bay leaf, cinnamon, and ginger rhizome on TNF-α serum levels of hyperglycemic rats.
Methods: The research design is used Laboratory experimental with control group post test only in vivo. Fourty male Wistar rats aged 2-3 months were divided into five groups: negative control group / KN (normal diet), positive control group / KP (High Fructose Diet / HFD and Streptozotocin / STZ), and three treatment groups. Combination infused was given by sonde to hyperglycemic groups for 30 days with dose 125 mg/kgBW, 250 mg/kgBW, 500 mg/kgBW respectively once daily. TNF-alpha serum levels was measured using ELISA (Enzyme Linked Immunosorbent-Assay) sandwich method and read by microplate reader with 450nm wavelength. The results were analyzed using One Way ANOVA statistical test.
Results: The given doses of combination of Sambiloto, bay leaf, cinnamon, and ginger rhizome unsignificantly decreased TNF-α serum level. The mean was found at KN (274 ± 11.15); KP (252 ± 14.79); KP1 (275.6 ± 5.93); KP2 (269.4 ± 18.58); KP3 (262 ± 9.2).
Conclusion: The combination doses of Sambiloto, bay leaf, cinnamon and ginger rhizome, 125mg/KgBW, 250mg/KgBW, 500mg/KgBW were not decrease TNF-alfa serum levels.
