Abstract


Introduction: Indomethacin increases the risk of developing gastric ulcers due to COX-1 inhibition. COX-1 inhibition decreases eNOS expression and induces an increase in COX-2 activity which will also increase iNOS expression. Excessive Nitric oxide (NO) will form excess peroxinitrite. Soursop leaf extract has the effect of being antioxidant and anti-inflammatory, which is thought to be able to prevent damage to the gastric mucosa. The aim of this study was to prove the effect of soursop leaf water extract to decrease the levels of NO rat gaster indomethacin induced.


Methods: The study was conducted in an experimental laboratory with control group post test only design. The experimental Animals are female wistar rats aged 10-12 weeks. They were divided into 5 groups: negative control group (KN, n = 5), positive control (KP, n = 5) with indomethacin dose 30 mg / KgBB, treatment group 1 (P1, n = 5) extract dose of 100mg / KgBB, treatment group 2 (P2, n = 5) extract dose 200 mg / KgBB, and treatment group 3 (P 3, n = 5) extract dose 400 mg / KgBB. Gastric NO levels were measured by the ELISA (Enzyme-linked immunosorbent assay) method to measure stable metabolites of nitrite and nitrate.


Results: Indomethacin significantly increased gastric NO levels (p <0.05). There was a tendency to decrease gastric NO levels with an increase in the extract dose given to the treatment group compared to KP. gastric NO levels of KN group did not differ significantly to P3 (p> 0.05).


Conclusion: Soursop leaves water extract has the potential effects to reduce levels of rat gastric NO that induced by indomethacin along with the increase in the dosage of the herb, close to normal levels.
