PENGARUH LAMA SIMPAN PROBIOTIK Lactobacillus salivarius TERENKAPSULASI ARABINOXYLAN TERHADAP TOTAL BAKTERI DAN JUMLAH BAKTERI ASAM LAKTAT
Abstract
Riset yang telah dilakukan memiliki tujuan untuk evaluasi pengaruh lama simpan probiotik L. salivarius dengan bahan penyalut (enkapsulasi) arabinoxylan terhadap total bakteri dan total koloni mikroba tipe bakteri asam laktat (BAL). Metode experimental dipilih dalam riset ini. Desain penelitian yang digunakan adalah Rancangan Acak Lengkap (RAL). Jumlah perlakuan sebanyak 4 dengan 5 kali ripetisi. Perlakuan yang digunakan yaitu P0 = penyimpanan hari ke-0, P1= penyimpanan hari ke-14, P2= penyimpanan hari ke-28, dan P3= penyimpanan hari ke-42. Analisis data pada riset ini adalah Analysis of Variance (ANOVA) dengan Uji Duncan jika diperoleh perbedaan antar perlakuan. Riset ini memberikan hasil berupa informasi terkait lama simpan berpengaruh sangat nyata (P<0,01) terhadap total bakteri dan jumlah BAL. Berdasarkan hasil penelitian didapatkan nilai total bakteri pada PO= 1,5 X 107, P1= 1,6 X 107, P2= 2,1 X 107, P3= 2,3 X 107. Hasil penelitian BAL diperoleh hasil P0= 2,4 X 107, P1= 2,5 X 107, P2= 2,1 X 107, P3= 1,5 X 107. Nilai total bakteri tertinggi diperoleh pada perlakuan P3 sebesar 2,3 × 107 cfu/g, sedangkan jumlah BAL terendah diperoleh pada perlakuan P3 sebesar 1,5 × 107 cfu/g. Berdasarkan hasil penelitian dapat disimpulkan bahwa lama simpan probiotik L. salivarius terenkapsulasi arabinoxylan berpengaruh sangat nyata terhadap total bakteri dan jumlah bakteri asam laktat. Lama simpan terbaik diperoleh pada penyimpanan hingga 14 hari karena pada periode tersebut nilai TPC masih rendah dan jumlah BAL masih tinggi sehingga kualitas probiotik masih baik.

EFFECT OF STORAGE DURATION OF ARABINOXYLAN ENCAPSULATED Lactobacillus salivarius PROBIOTIC ON TOTAL BACTERIAL COUNT AND LACTIC ACID BACTERIA COUNT
Abstract
This study aimed to evaluate the effect of the storage period of arabinoxylan-encapsulated Lactobacillus salivarius probiotic on the total bacterial count and the population of lactic acid bacteria (LAB). An experimental method was employed using a Completely Randomized Design (CRD) consisting of four treatments with five replications. The treatments were as follows: P0 = 0 days of storage, P1 = 14 days of storage, P2 = 28 days of storage, and P3 = 42 days of storage. Data were analyzed using Analysis of Variance (ANOVA), followed by Duncan's Multiple Range Test when significant differences among treatments were detected. The results showed that the storage period had a highly significant effect (P < 0.01) on both the total bacterial count and the LAB population. The total bacterial counts were 1.5 × 10⁷, 1.6 × 10⁷, 2.1 × 10⁷, and 2.3 × 10⁷ CFU/g for P0, P1, P2, and P3, respectively. Meanwhile, the LAB counts were 2.4 × 10⁷, 2.5 × 10⁷, 2.1 × 10⁷, and 1.5 × 10⁷ CFU/g for P0, P1, P2, and P3, respectively. The highest total bacterial count was observed in P3 (2.3 × 10⁷ CFU/g), whereas the lowest LAB count was recorded in P3 (1.5 × 10⁷ CFU/g). In conclusion, the storage period of arabinoxylan-encapsulated L. salivarius probiotic had a highly significant effect on both the total bacterial count and the LAB population. The optimum storage period was 14 days, as the Total Plate Count (TPC) remained relatively low while the LAB population remained high, indicating that the probiotic quality was still well maintained.
