PENGARUH JENIS KEMASAN TERHADAP TOTAL KOLONI BAKTERI DAN TOTAL KOLONI BAKTERI ASAM LAKTAT DARI PROBIOTIK Lactobacillus salivarius TERENKAPSULASI ARABINOXYLAN
Abstract
Penelitian ini bertujuan untuk mengetahui pengaruh berbagai jenis kemasan terhadap Total Koloni Bakteri dan Total Koloni Bakteri Asam Laktat dari probiotik Lactobacillus salivarius yang terenkapsulasi arabinoxylan selama masa penyimpanan. Metode penelitian yang digunakan adalah metode eksperimen dengan Rancangan Acak Lengkap (RAL) yang terdiri dari 4 perlakuan kemasan (P0: tanpa kemasan, P1: aluminium foil, P2: kardus, P3: kombinasi aluminium foil dan kardus) dengan lima ulangan. Analisis data dilakukan menggunakan Analysis of Variance (ANOVA) dan dilanjutkan dengan menggunakan aplikasi SPSS 16 Uji Duncan Multiple Range Test (DMRT) jika terdapat pengaruh yang nyata. Hasil analisis statistik menunjukkan bahwa jenis kemasan tidak memberikan pengaruh nyata (P≥0,05) terhadap Total Koloni Bakteri, di mana jumlah populasi mikroba heterotrof pada seluruh perlakuan cenderung stabil dan seragam. Sebaliknya, jenis kemasan memberikan pengaruh yang nyata (P≤0,05) terhadap Total Koloni Bakteri Asam Laktat selama periode penyimpanan 30 hari. Jumlah Total Koloni Bakteri Asam Laktat tertinggi ditemukan pada kemasan kardus (P2) sebesar 1,5 x 105 CFU, sedangkan jumlah terendah terdapat pada kemasan aluminium foil (P1) sebesar 7,7 × 107 CFU. Karakteristik kemasan kardus yang porus diduga mampu menjaga keseimbangan difusi gas hasil metabolisme dan melindungi sel dari fluktuasi suhu secara optimal. Kesimpulan dari penelitian ini adalah jenis kemasan terbukti memengaruhi Total Koloni Bakteri Asam Laktat tetapi tidak memengaruhi Total Koloni Bakteri, dengan kemasan kardus sebagai media penyimpanan yang paling efektif.

THE EFFECT OF PACKAGING TYPE ON TOTAL BACTERIAL COLONIES AND TOTAL LACTIC ACID BACTERIA COLONIES OF ENCAPSULATED PROBIOTIC Lactobacillus salivarius WITH ARABINOXYLAN
Abstract
This study aimed to determine the effect of various packaging types on the Total Bacterial Colonies and Total Lactic Acid Bacteria colonies of Lactobacillus salivarius probiotics encapsulated with arabinoxylan during storage. The research method utilized an experimental approach with a Completely Randomized Design (CRD), consisting of four packaging treatments (P0: no packaging, P1: aluminum foil, P2: cardboard, P3: combination of aluminum foil and cardboard) with five replications. Data were analyzed using Analysis of Variance (ANOVA) and, if a significant effect was detected, further tested using the Duncan Multiple Range Test (DMRT) with the SPSS 16 application. Statistical analysis showed that the packaging type had no significant effect (P≥0,05) on Total Bacterial Colonies, where the heterotrophic microbial population across all treatments tended to be stable and uniform. Conversely, the packaging type significantly affected (P≤0,05) the Total Lactic Acid Bacteria Colonies during the 30-day storage period. The highest Total Lactic Acid Bacteria Colonies count was found in cardboard packaging (P2) at 1.5 x 105 CFU, while the lowest count was observed in aluminum foil packaging (P1) at 7,7 × 107 CFU. The porous characteristic of cardboard packaging is suggested to effectively maintain the balance of metabolic gas diffusion and optimally protect the cells from temperature fluctuations. In conclusion, the packaging type significantly influences the of Total Lactic Acid Bacteria Colonies but does not affect total bacterial colonies, with cardboard packaging being the most effective storage medium.
